Partition test and sexual motivation in male mice

Abstract

Theoretical analysis of own and literature investigations of sexual motivation with the use of the partition test [Kudryavtseva, 1987, 1994] in male mice was carried out. It has been shown that appearance of a receptive female in the neighboring compartment of common cage separated by perforated transparent partition produces the enhancement of testosterone level in blood and stimulates the behavioral activity near partition as a reaction to the receptive female in naive males. In many studies this behavioral activity is considered as sexual motivation, arising in this experimental context in male mice. The lack of correlation between behavioral parameters and gonad reaction of males on receptive female, uninterconnected changes of these two parameters as well as the lack of sexual behavior between naive male and female when partition is removed cast doubt on this data interpretation. It has been supposed that in naive males behavioral reaction to a receptive female is induced by positive incentive - odor of the female associated with nursing and warmth from mother and other females which look after posterity. Short-term increase of the level of testosterone (possessing rewarding properties) is innate stimulus-response reaction which stimulates and prolongs behavioral interest of male to receptive female. It has been supposed that after sexual experience female odor is associated in experienced males with sexual behavior directed to the sexual partner and resulted in the formation of sexual motivation. The data are considered also in the light of the theory of motivated behavior including "liking", "wanting" and "learning" [Robinson and Berridge, 1993, 2000].

Drug-Associated Rhabdomyolysis in the FDA Adverse Event Reporting System (FAERS): A Disproportionality Analysis

This study aimed to systematically identify and quantify risks for drug-induced rhabdomyolysis (DIR) using real-world data and to propose an evidence-based risk mitigation framework. We conducted a retrospective pharmacovigilance study using the FDA Adverse Event Reporting System (FAERS) database from Q1 2005 to Q1 2025. A two-stage analysis involved initial signal detection using the Reporting Odds Ratio (ROR), followed by a LASSO-optimized multivariate logistic regression to calculate adjusted odds ratios (aORs) for 54 target drugs while controlling for confounders. Our analysis confirmed potent DIR risks for known agents, such as gemfibrozil (aOR 173.67) and statins (lovastatin aOR 97.20, simvastatin aOR 85.12). Crucially, we identified strong, novel risk signals for drugs currently lacking warnings, most notably levetiracetam (aOR 11.02) and donepezil (aOR 8.90). A significant "labeling gap" was quantified: 61.1% of drugs with a statistically significant DIR risk lack a corresponding warning in U.S. drug labels. We subsequently developed a three-tiered risk stratification model. The proposed framework provides a data-driven foundation for developing tiered clinical decision support systems, enhancing prescribing safety, and guiding future regulatory action to bridge the identified evidence-to-labeling gap.

Current validation practice undermines surgical AI development

Surgical data science (SDS) is rapidly advancing, yet clinical adoption of artificial intelligence (AI) in surgery remains limited, with inadequate validation as an important contributing factor. Existing validation practices often neglect the temporal and hierarchical structure of intraoperative videos, yielding misleading or clinically irrelevant results. We introduce a comprehensive catalogue of validation pitfalls in AI-based surgical video analysis, derived from a multi-stage Delphi process with 92 international experts. Pitfalls span three categories: (1) data, (2) metric selection/configuration, and (3) aggregation and reporting. A systematic review of surgical AI papers reveals that these pitfalls are widespread. Experiments on surgical video datasets show that ignoring temporal and hierarchical data structures can understate uncertainty, obscure critical failure modes, and alter algorithm rankings. To address these shortcomings, we provide consensus-based best practices compiled. Together, this work provides an evidence-based framework for rigorous validation of surgical video analysis algorithms, guiding benchmarking, reporting, regulatory review, and clinical translation.

Cancer genomics, targeted therapy to precision oncology: the treatment paradigm is a holistic approach to cancer therapeutics

Cancer is a fatal genetic disease involving unregulated cell growth and proliferation. Cellular behaviors such as growth, proliferation, survival, or death rely upon the ability of cells to communicate with the surrounding system; therefore, effective targeting of dysregulated signaling pathways involving growth factors, regulatory proteins, cell adhesion molecules, and immune system molecules is critical to cancer therapy. They are mainly driven by alterations in tumor suppressor genes and oncogenes that may vary among different cancer types. Importantly, patients with the same cancer type respond differently to available cancer treatments, likely due to tumor-specific DNA, RNA, and proteins, indicating the need for patient-specific treatment options. Precision oncology has evolved as a form of cancer therapy focused on genetic and molecular profiling of tumors to identify specific molecular alterations involved in carcinogenesis for tailored individualized cancer treatment. This article aims to briefly explain the foundations and frontiers of precision oncology, highlighting the cutting-edge innovations in tools and techniques associated with it to assess its scope and importance in achieving efficacious cancer treatment over time.
