Mining for Potent Inhibitors through Artificial Intelligence and Physics: A Unified Methodology for Ligand Based and Structure Based Drug Design

The viability of a new drug molecule is a time and resource intensive task that makes computer-aided assessments a vital approach to rapid drug discovery. Here we develop a machine learning algorithm, iMiner, that generates novel inhibitor molecules for target proteins by combining deep reinforcement learning with real-time 3D molecular docking using AutoDock Vina, thereby simultaneously creating chemical novelty while constraining molecules for shape and molecular compatibility with target active sites. Moreover, through the use of various types of reward functions, we can generate new molecules that are chemically similar to a target ligand, which can be grown from known protein bound fragments, as well as to create molecules that enforce interactions with target residues in the protein active site. The iMiner algorithm is embedded in a composite workflow that filters out Pan-assay interference compounds, Lipinski rule violations, and poor synthetic accessibility, with options for cross-validation against other docking scoring functions and automation of a molecular dynamics simulation to measure pose stability. Because our approach only relies on the structure of the target protein, iMiner can be easily adapted for future development of other inhibitors or small molecule therapeutics of any target protein.

Progress and Prospects in 3D Generative AI: A Technical Overview including 3D human

While AI-generated text and 2D images continue to expand its territory, 3D generation has gradually emerged as a trend that cannot be ignored. Since the year 2023 an abundant amount of research papers has emerged in the domain of 3D generation. This growth encompasses not just the creation of 3D objects, but also the rapid development of 3D character and motion generation. Several key factors contribute to this progress. The enhanced fidelity in stable diffusion, coupled with control methods that ensure multi-view consistency, and realistic human models like SMPL-X, contribute synergistically to the production of 3D models with remarkable consistency and near-realistic appearances. The advancements in neural network-based 3D storing and rendering models, such as Neural Radiance Fields (NeRF) and 3D Gaussian Splatting (3DGS), have accelerated the efficiency and realism of neural rendered models. Furthermore, the multimodality capabilities of large language models have enabled language inputs to transcend into human motion outputs. This paper aims to provide a comprehensive overview and summary of the relevant papers published mostly during the latter half year of 2023. It will begin by discussing the AI generated object models in 3D, followed by the generated 3D human models, and finally, the generated 3D human motions, culminating in a conclusive summary and a vision for the future.

DeLR: Active Learning for Detection with Decoupled Localization and Recognition Query

Active learning has been demonstrated effective to reduce labeling cost, while most progress has been designed for image recognition, there still lacks instance-level active learning for object detection. In this paper, we rethink two key components, i.e., localization and recognition, for object detection, and find that the correctness of them are highly related, therefore, it is not necessary to annotate both boxes and classes if we are given pseudo annotations provided with the trained model. Motivated by this, we propose an efficient query strategy, termed as DeLR, that Decoupling the Localization and Recognition for active query. In this way, we are probably free of class annotations when the localization is correct, and able to assign the labeling budget for more informative samples. There are two main differences in DeLR: 1) Unlike previous methods mostly focus on image-level annotations, where the queried samples are selected and exhausted annotated. In DeLR, the query is based on region-level, and we only annotate the object region that is queried; 2) Instead of directly providing both localization and recognition annotations, we separately query the two components, and thus reduce the recognition budget with the pseudo class labels provided by the model. Experiments on several benchmarks demonstrate its superiority. We hope our proposed query strategy would shed light on researches in active learning in object detection.

Visualization of Oxygen Vacancies and Self-doped Ligand Holes in La3Ni2O7-δ

The recent discovery of superconductivity in La3Ni2O7-δ under high pressure with a transition temperature around 80 K has sparked extensive experimental and theoretical efforts. Several key questions regarding the pairing mechanism remain to be answered, such as the most relevant atomic orbitals and the role of atomic deficiencies. Here, we develop a new energy-filtered multislice electron ptychography technique, assisted with electron energy loss spectroscopy, to address these critical issues. Oxygen vacancies are directly visualized and are found to primarily occupy the inner apical sites, which have been proposed to be crucial to superconductivity. We precisely determine the nanoscale stoichiometry and its correlation to the oxygen K edge spectra, which reveals a significant inhomogeneity in the oxygen content and electronic structure within the sample. The spectroscopic results also unveil that stoichiometric La3Ni2O7 is in the charge-transfer regime, with holes that are self-doped from Ni sites into O sites. The outer apical oxygen is found to be less relevant to the low-energy physics and can be safely disregarded in theoretical models. These observations will assist in further development and understanding of superconducting nickelate materials. Our imaging technique for quantifying atomic deficiencies can also be widely applied in materials science and condensed matter physics.

Pressure-dependent "Insulator-Metal-Insulator" Behavior in Sr-doped La$_3$Ni$_2$O$_7$

Recently, superconductivity at high temperatures has been observed in bulk La$_3$Ni$_2$O$_{7-δ}$ under high pressure. However, the attainment of high-purity La$_3$Ni$_2$O$_{7-δ}$ single crystals, exhibiting controlled and homogeneous stoichiometry through the post-annealing process in an oxygen-rich floating zone furnace, remains a formidable challenge. Here, we report the crystal structure and physical properties of single crystals of Sr-doped La$_3$Ni$_2$O$_7$ synthesized at high pressure (20 GPa) and high temperature (1400 °C). Through single crystal X-ray diffraction, we showed that high-pressure-synthesized paramagnetic Sr-doped La$_3$Ni$_2$O$_7$ crystallizes in an orthorhombic structure with Ni-O-Ni bond angles of 173.4(2)°out-of-plane and 175.0(2)°and 176.7(2)°in plane. The substitution of Sr alters in band filling and the ratio of Ni$^{2+}$/Ni$^{3+}$ in Sr-doped La$_3$Ni$_2$O$_7$, aligning them with those of "La$_3$Ni$_2$O$_{7.05}$", thereby leading to significant modifications in properties under high pressure relative to the unsubstituted parent phase. At ambient pressure, Sr-doped La$_3$Ni$_2$O$_7$ exhibits insulating properties, and the conductivity increases as pressure goes up to 10 GPa. However, upon further increasing pressure beyond 10.7 GPa, Sr-doped La$_3$Ni$_2$O$_7$ transits back from a metal-like behavior to an insulator. The insulator-metal-insulator trend under high pressure dramatically differs from the behavior of the parent compound La$_3$Ni$_2$O$_{7-δ}$, despite their similar behavior in the low-pressure regime. These experimental results underscore the considerable challenge in achieving superconductivity in nickelates.

Strong squeezing of microwave output fields via reservoir-engineered cavity magnomechanics

We show how to achieve strong squeezing of a microwave output field by reservoir engineering a cavity magnomechanical system, consisting of a microwave cavity, a magnon mode, and a mechanical vibration mode. The magnon mode is simultaneously driven by two microwave fields at the blue and red sidebands associated with the vibration mode. The two-tone drive induces a squeezed magnonic reservoir for the intracavity field, leading to a squeezed cavity mode due to the cavity-magnon state swapping, which further yields a squeezed cavity output field. The squeezing of the output field is stationary and substantial using currently available parameters in cavity magnomechanics. The work indicates the potential of the cavity magnomechanical system in preparing squeezed microwave fields, and may find promising applications in quantum information science and quantum metrology.

A Dual Domain Multi-exposure Image Fusion Network based on the Spatial-Frequency Integration

Multi-exposure image fusion aims to generate a single high-dynamic image by integrating images with different exposures. Existing deep learning-based multi-exposure image fusion methods primarily focus on spatial domain fusion, neglecting the global modeling ability of the frequency domain. To effectively leverage the global illumination modeling ability of the frequency domain, we propose a novelty perspective on multi-exposure image fusion via the Spatial-Frequency Integration Framework, named MEF-SFI. Initially, we revisit the properties of the Fourier transform on the 2D image, and verify the feasibility of multi-exposure image fusion on the frequency domain where the amplitude and phase component is able to guide the integration of the illumination information. Subsequently, we present the deep Fourier-based multi-exposure image fusion framework, which consists of a spatial path and frequency path for local and global modeling separately. Specifically, we introduce a Spatial-Frequency Fusion Block to facilitate efficient interaction between dual domains and capture complementary information from input images with different exposures. Finally, we combine a dual domain loss function to ensure the retention of complementary information in both the spatial and frequency domains. Extensive experiments on the PQA-MEF dataset demonstrate that our method achieves visual-appealing fusion results against state-of-the-art multi-exposure image fusion approaches. Multi-Scene Generalized Trajectory Global Graph Solver with Composite Nodes for Multiple Object Tracking

The global multi-object tracking (MOT) system can consider interaction, occlusion, and other ``visual blur'' scenarios to ensure effective object tracking in long videos. Among them, graph-based tracking-by-detection paradigms achieve surprising performance. However, their fully-connected nature poses storage space requirements that challenge algorithm handling long videos. Currently, commonly used methods are still generated trajectories by building one-forward associations across frames. Such matches produced under the guidance of first-order similarity information may not be optimal from a longer-time perspective. Moreover, they often lack an end-to-end scheme for correcting mismatches. This paper proposes the Composite Node Message Passing Network (CoNo-Link), a multi-scene generalized framework for modeling ultra-long frames information for association. CoNo-Link's solution is a low-storage overhead method for building constrained connected graphs. In addition to the previous method of treating objects as nodes, the network innovatively treats object trajectories as nodes for information interaction, improving the graph neural network's feature representation capability. Specifically, we formulate the graph-building problem as a top-k selection task for some reliable objects or trajectories. Our model can learn better predictions on longer-time scales by adding composite nodes. As a result, our method outperforms the state-of-the-art in several commonly used datasets.

Impersonating a Superconductor: High-Pressure BaCoO$_3$, an Insulating Ferromagnet

We report the high-pressure synthesis (6 GPa, 1200 $^{\circ}$C) and ambient pressure characterization of hexagonal HP-BaCoO$_3$. The material (with the 2H crystal structure) has a short intrachain Co-Co distance of about 2.07 $\text{Å}$. Our magnetization investigation revealed robust diamagnetic behavior below approximately 130 K when exposed to weak applied magnetic fields (10 Oe) and a distinct half-levitation phenomenon below that temperature, such as is often observed for superconductors. Its field-dependent magnetization profile, however, unveils the characteristics of ferromagnetism, marked by a substantial magnetic retentivity of 0.22(1) $μ_B$/Co at a temperature of 2 K. Electrical resistivity measurements indicate that HP-BaCoO$_3$ is a ferromagnetic insulator, not a superconductor.

6G Network AI Architecture for Everyone-Centric Customized Services

Mobile communication standards were developed for enhancing transmission and network performance by using more radio resources and improving spectrum and energy efficiency. How to effectively address diverse user requirements and guarantee everyone's Quality of Experience (QoE) remains an open problem. The Sixth Generation (6G) mobile systems will solve this problem by utilizing heterogenous network resources and pervasive intelligence to support everyone-centric customized services anywhere and anytime. In this article, we first coin the concept of Service Requirement Zone (SRZ) on the user side to characterize and visualize the integrated service requirements and preferences of specific tasks of individual users. On the system side, we further introduce the concept of User Satisfaction Ratio (USR) to evaluate the system's overall service ability of satisfying a variety of tasks with different SRZs. Then, we propose a network Artificial Intelligence (AI) architecture with integrated network resources and pervasive AI capabilities for supporting customized services with guaranteed QoEs. Finally, extensive simulations show that the proposed network AI architecture can consistently offer a higher USR performance than the cloud AI and edge AI architectures with respect to different task scheduling algorithms, random service requirements, and dynamic network conditions.

HODN: Disentangling Human-Object Feature for HOI Detection

The task of Human-Object Interaction (HOI) detection is to detect humans and their interactions with surrounding objects, where transformer-based methods show dominant advances currently. However, these methods ignore the relationship among humans, objects, and interactions: 1) human features are more contributive than object ones to interaction prediction; 2) interactive information disturbs the detection of objects but helps human detection. In this paper, we propose a Human and Object Disentangling Network (HODN) to model the HOI relationships explicitly, where humans and objects are first detected by two disentangling decoders independently and then processed by an interaction decoder. Considering that human features are more contributive to interaction, we propose a Human-Guide Linking method to make sure the interaction decoder focuses on the human-centric regions with human features as the positional embeddings. To handle the opposite influences of interactions on humans and objects, we propose a Stop-Gradient Mechanism to stop interaction gradients from optimizing the object detection but to allow them to optimize the human detection. Our proposed method achieves competitive performance on both the V-COCO and the HICO-Det datasets. It can be combined with existing methods easily for state-of-the-art results.

Real-Space Visualization of Frequency-Dependent Anisotropy of Atomic Vibrations

The underlying dielectric properties of materials, intertwined with intriguing phenomena such as topological polariton modes and anisotropic thermal conductivities, stem from the anisotropy in atomic vibrations. Conventionally, X-ray diffraction techniques have been employed to estimate thermal ellipsoids of distinct elements, albeit lacking the desired spatial and energy resolutions. Here we introduce a novel approach utilizing the dark-field monochromated electron energy-loss spectroscopy for momentum-selective vibrational spectroscopy, enabling the cartographic delineation of variations of phonon polarization vectors. By applying this technique to centrosymmetric cubic-phase strontium titanate, we successfully discern two types of oxygen atoms exhibiting contrasting vibrational anisotropies below and above 60 meV due to their frequency-linked thermal ellipsoids. This method establishes a new pathway to visualize phonon eigenvectors at specific crystalline sites for diverse elements, thus delving into uncharted realms of dielectric, optical, and thermal property investigations with unprecedented spatial resolutions.

Magnon squeezing by two-tone driving of a qubit in cavity-magnon-qubit systems

We propose a scheme for preparing magnon squeezed states in a hybrid cavity-magnon-qubit system. The system consists of a microwave cavity that simultaneously couples to a magnon mode of a macroscopic yttrium-iron-garnet (YIG) sphere via the magnetic-dipole interaction and to a transmon-type superconducting qubit via the electric-dipole interaction. By far detuning from the magnon-qubit system, the microwave cavity is adiabatically eliminated. The magnon mode and the qubit then get effectively coupled via the mediation of virtual photons of the microwave cavity. We show that by driving the qubit with two microwave fields and by appropriately choosing the drive frequencies and strengths, magnonic parametric amplification can be realized, which leads to magnon quadrature squeezing with the noise below vacuum fluctuation. We provide optimal conditions for achieving magnon squeezing, and moderate squeezing can be obtained using currently available parameters. The generated squeezed states are of a magnon mode involving more than $10^{18}$ spins and thus macroscopic quantum states. The work may find promising applications in quantum information processing and high-precision measurements based on magnons and in the study of macroscopic quantum states.

Synchronization by Magnetostriction

We show how to utilize magnetostriction to synchronize two mechanical vibration modes in a cavity magnomechanical system. The dispersive magnetostrictive interaction provides necessary nonlinearity required for achieving synchronization. Strong phase correlation between two mechanical oscillators can be established, leading to the synchronization robust against thermal noise. We develop a theoretical framework to analyze the synchronization by solving the constraint conditions of steady-state limit cycles. We determine that the strong cavity-magnon linear coupling can enhance and regulate the synchronization, which offers a new path to modulate synchronization. The work reveals a new mechanism for achieving and modulating synchronization and indicates that cavity magnomechanical systems can be an ideal platform to explore rich synchronization phenomena.

Audio-visual Saliency for Omnidirectional Videos

Visual saliency prediction for omnidirectional videos (ODVs) has shown great significance and necessity for omnidirectional videos to help ODV coding, ODV transmission, ODV rendering, etc.. However, most studies only consider visual information for ODV saliency prediction while audio is rarely considered despite its significant influence on the viewing behavior of ODV. This is mainly due to the lack of large-scale audio-visual ODV datasets and corresponding analysis. Thus, in this paper, we first establish the largest audio-visual saliency dataset for omnidirectional videos (AVS-ODV), which comprises the omnidirectional videos, audios, and corresponding captured eye-tracking data for three video sound modalities including mute, mono, and ambisonics. Then we analyze the visual attention behavior of the observers under various omnidirectional audio modalities and visual scenes based on the AVS-ODV dataset. Furthermore, we compare the performance of several state-of-the-art saliency prediction models on the AVS-ODV dataset and construct a new benchmark. Our AVS-ODV datasets and the benchmark will be released to facilitate future research.

Augmenting Lane Perception and Topology Understanding with Standard Definition Navigation Maps

Autonomous driving has traditionally relied heavily on costly and labor-intensive High Definition (HD) maps, hindering scalability. In contrast, Standard Definition (SD) maps are more affordable and have worldwide coverage, offering a scalable alternative. In this work, we systematically explore the effect of SD maps for real-time lane-topology understanding. We propose a novel framework to integrate SD maps into online map prediction and propose a Transformer-based encoder, SD Map Encoder Representations from transFormers, to leverage priors in SD maps for the lane-topology prediction task. This enhancement consistently and significantly boosts (by up to 60%) lane detection and topology prediction on current state-of-the-art online map prediction methods without bells and whistles and can be immediately incorporated into any Transformer-based lane-topology method. Realization of the unidirectional amplification in a cavity magnonic system

We experimentally demonstrate the nonreciprocal microwave amplification using a cavity magnonic system, consisting of a passive cavity (i.e., the split-ring resonator), an active feedback circuit integrated with an amplifier, and a ferromagnetic spin ensemble (i.e., a yttrium-iron-garnet sphere). Combining the amplification provided by the active circuit and the nonreciprocity supported by the cavity magnonics, we implement a nonreciprocal amplifier with the functions of both unidirectional amplification and reverse isolation. The microwave signal is amplified by 11.5 dB in the forward propagating direction and attenuated in the reverse direction by -34.7 dB, giving an isolation ratio of 46.2 dB. Such a unidirectional amplifier can be readily employed in quantum technologies, where the device can simultaneously amplify the weak signal output by the quantum system and isolate the sensitive quantum system from the backscattered external noise. Also, it is promising to explore more functions and applications using a cavity magnonic system with real gain.

Efficient Serverless Function Scheduling at the Network Edge

Serverless computing is a promising approach for edge computing since its inherent features, e.g., lightweight virtualization, rapid scalability, and economic efficiency. However, previous studies have not studied well the issues of significant cold start latency and highly dynamic workloads in serverless function scheduling, which are exacerbated at the resource-limited network edge. In this paper, we formulate the Serverless Function Scheduling (SFS) problem for resource-limited edge computing, aiming to minimize the average response time. To efficiently solve this intractable scheduling problem, we first consider a simplified offline form of the problem and design a polynomial-time optimal scheduling algorithm based on each function's weight. Furthermore, we propose an Enhanced Shortest Function First (ESFF) algorithm, in which the function weight represents the scheduling urgency. To avoid frequent cold starts, ESFF selectively decides the initialization of new function instances when receiving requests. To deal with dynamic workloads, ESFF judiciously replaces serverless functions based on the function weight at the completion time of requests. Extensive simulations based on real-world serverless request traces are conducted, and the results show that ESFF consistently and substantially outperforms existing baselines under different settings.
